Proteomics

This is the new field that has emerged in the post-genome era.  Proteomics includes the research area of proteins expressed from genes or proteins in general. The 2D electrophoresis followed by image analysis and further qualitatively analyzed by mass spectrometry is the general strategy accepted by many researchers now.

Microarray

A new method for the detection of thousands of RNA at one time. RNA in the sample are processed for reverse transcription and fluorescently labeled. After hybridization of the fluorescently labeled cDNA and the microarray slide, image analysis of the glass slide is done by a high-resolution fluorescence detector.

Southern Blotting

A widely used method for the detection of a specific sequence in DNA. This method was named after Dr. E.M. Southern who introduced this method in 1975. After the gel electrophoresis of the sample DNA, blotting to a membrane is done and DNAs are transcribed to the membrane. A DNA probe of specific sequence is hybridized with the membrane. When the probe is a radioisotope (RI), exposure to phosphor imaging plate (IP) and detection by BAS system is done. When the probe is digoxigenine (DIG), a sandwich method using enzyme (ALP or HRP) tagged anti-DIG-antibody and chemiluminescent detection by using chemiluminescence substrate is done. High sensitive detection of chemiluminescence is effectively done by a cooled CCD camera system.

Western Blotting

A widely used method for the detection of specific protein after electrophoresis. The sample is electrophoresed on a polyacrylamide gel then blotted to a membrane. The membrane is incubated with the antibody to the specific protein. Further, the secondary antibody labeled with an enzyme such as HRP (or ALP) is added and reacted. Chemiluminescence detection of the specific protein is done by applying chemiluminescent substrate of the enzyme.

Northern Blotting

A widely used method for detection of a specific RNA after electrophoresis. The electrophoresis gel is blotted to a membrane and the transcribed RNA is detected by hybridization with a labeled cDNA probe. Phosphor imaging plate (IP) is used for the detection of RI probes, and very high sensitive cooled CCD camera system is used for detection of chemiluminescent probes.  Because we can get molecular weight information from the electrophoresis length, Northern blotting is useful for verification purposes.

GFP (Green Fluorescent Protein)

A new method for visualizing various processes of metabolism in a cell. Fluorescent scanner or CCD camera system with appropriate excitation light source can be used for the imaging of GFP.
